(A) Time course of ATP-dependent relaxation supported by A59₂B₂. The reaction contained 3.1 nM enzyme and 3.5 nM supercoiled pBR322 and was incubated at 25°C for the times indicated. Samples were analyzed on a 1% agarose gel (o.c., open circular; s/c, negatively supercoiled; rel., relaxed). (B) Inhibition of the relaxation of supercoiled pBR322 (3.5 nM) with A59₂B₂ (62 nM) by the addition of 100 μM novobiocin after 10 min.
