Effect of mutations of the rasiRNA pathway genes on the presence of sense and antisense HeT-A-specific rasiRNAs in ovaries. Northern analysis of the RNA isolated from ovaries of spn-E1/TM3, spn-E1/spn-Ehls3987, piwi2/CyO, piwi2/piwi3, aubQC42/CyO and aubQC42/aubHN flies. +/− and −/− designate hetero- and transheterozygous states of the corresponding mutations, respectively. Hybridization was done with HeT-A riboprobes from the HeT-A 3′-region to detect antisense (left part) or sense (right part) HeT-A-specific rasiRNAs. Lower panels represent hybridization with oligonucleotide complementary to the mir-13b1 microRNA or 2S RNA. P33-labeled RNA oligonucleotides were used as size markers.
