Triton X-114 phase partitioning to assess the hydrophobicity of palmitoylated and unpalmitoylated α_s. Membrane-derived palmitoylated α_s-WT incubated without or with palmitoyl esterase, α_s-C3S, cytosol-derived α_s-WT, or βγ subunit (each at 50 nM) were subjected to TX-114 phase partitioning as described. Equal volumes of normalized fractions of either water phase (W) or detergent phase (D) were subjected to SDS/PAGE and visualized either by immunoblotting with anti-α_s or anti-β antibody or by fluorography.
