Effects of Rsf-D4 expression on OVCAR3 ovarian cancer cells. A: In a co-immunoprecipitation assay, recombinant Rsf-D4 proteins show a does-dependent inhibition in the binding of Rsf-1 and hSNF2H. GAPDH was used as a loading control. B: Western blotting demonstrates a robust Rsf-1 protein expression in OVCAR3 cell line and a moderate expression in the A2780 cell line as compared to other ovarian carcinoma cell lines. C: The effects of deletion mutants were assessed by comparing the cell number of the OVCAR3 cells after transient transfection of Rsf-1 to Rsf-10 constructs. The cell number was measured at day 4 using SYBR green I incorporation and the data were normalized to the empty vector control. D: The cellular proliferating activity in the transfected OVCAR3 cells was determined by the percentage of cells with positive BrdU staining. E: The percentage of apoptotic cells in each treatment was determined by annexin V-FITC staining.
