Respiratory enzyme and ATP levels in lentiviral transductants. A, levels of respiratory complexes I and IV in total cell extracts were quantitated by dipstick assay (MitoSciences). The top band represents the positive control signal; the bottom band represents the specific signal from the respective complexes. Membranes were scanned for densitometric analysis, and the percentage of enzyme per unit of protein in the PRC shRNA1 transductant relative to the control shRNA is expressed as the average ± S.E. for three separate determinations. B, steady-state ATP levels were quantitated in total cell extracts using a bioluminescent assay. Values for oligomycin-sensitive ATP levels for control shRNA, PRC shRNA1, and PRC shRNA4 transductants are shown as the average ± S.E. for three separate determinations.
