Table 2.

Mean association and dissociation kinetic rate constants (kon, koff) and equilibrium dissociation constants (KD), calculated from surface plasmon resonance (SPR) measurements for aqueous Congo red (CR) flowing past chip-linked FP and Aβ(26–42) monolayersa

Chip-linked peptide kon (1/Ms) koff (1/s) KD (nM)
FP 62.9 × 105 18.7 × 10−3 2.97
Aβ(26–42) 19.2 × 105 4.0 × 10−3 2.10

Congo red in HBS-EP buffer was flowed past monolayers of FP or Aβ(26–42), linked via their respective N-terminal amides to CM5 sensor chips in a Biacore 3000 system (see section 2). Mean kinetic rate constants (kon, koff) and equilibrium dissociation constants (KD = koff/kon) were determined from curve fitting analyses of SPR results at 10 nM CR.
