Quantification of Insoluble and Soluble Glucans in the Wild Type, isa1/isa2, the Quadruple DBE Mutant, and the Quintuple Mutant Lacking All Four DBEs and the Endoamylase AMY3.
Plants were harvested and immediately frozen in liquid N2. Each sample comprised a complete individual rosette. Total insoluble and soluble glucans were extracted and measured as in Figure 1. Wild-type and mutant plants were harvested at the end of a 12-h photoperiod (white bars) and at the end of the night (black bars). Each value is the mean ± se of four replicate samples. Note the increase in glucan content of the quintuple mutant. A second experiment performed with an independently grown batch of plants yielded similar results.
