Western blot analysis for AP-1 components and upstream modulators. Western blot analyses were performed using 30 μg total cell protein extracted from K12 and K7M2 osteosarcoma cells separated by SDS-polyacrylamide gel electrophoresis. After transfer of the proteins to nitrocellulose membranes the blots were probed with antibodies against cJun, p-cJun, JunB, JunD, cFos, Fra1, ATF2, ATF3, JIP1, JAB1, and β-tubulin as a control for protein loading. All blots were generated using the same protein extract, with the p-cJun blots stripped to analyze total cJun levels. The results shown are representative of three independent experiments.
