Effects of selective caspase inhibition on LCA- and
ent-LCA-induced caspase activation in HT-29 cells. Selective
inhibitors of caspase-2 and caspase-8, a pan-caspase inhibitor, or vehicle
control (CTRL, Me2SO) were used as pretreatment for HT-29
cells treated with 500 μm LCA for 1 h (A) and HT-29
cells treated with 500 μm ent-LCA for 1 h (B).
In all of the experiments caspase inhibitor treated cells were pretreated with
50 μm Z-VDVAD-FMK (2 indicates caspase-2 inhibitor),
Z-IETD-FMK (8 indicates caspase-8 inhibitor), or Z-VAD-FMK
(Pan indicates pan-caspase inhibitor) for 4 h prior to bile acid
exposure.
