PKCλ/ι is up-regulated upon T-cell activation and differentiation and is required for Th2-cytokine secretion. (A) Purified naïve splenic CD4+ T cells or cells from cervical, axillary, inguinal, and popliteal lymph nodes (LN) were stimulated with anti-CD3/CD28 for 24 or 48 hours. (B) Splenic CD4+ T were differentiated under Th0, Th1, and Th2 conditions for 4 days. Cells in A and B were restimulated with anti-CD3/CD28 for 2 days. Cells were collected for Western blot analysis. Results shown are from a single experiment representative of 2 independent experiments. (C and D) Purified naïve splenic CD4+ T cells were stimulated with (C) anti-CD3/CD28 for 3 days or (D) differentiated under Th0/Th1/Th2 conditions for 4 days and restimulated with plate-bound anti-CD3 for 1 day.
