(A) Immunlocalization of MMP-2, MMP-9 and MT1-MMP in serial paraffin cross-sections of chicken embryo aorta at days E7 and E14 at day 7, a moderate immunoreactivity for MMP-2 and MMP-9 is detected in the endothelium (e), condensed mesenchyme (cm) and lamellar cell layers (lc). Less intense and more restricted immunoreactivity for MT1-MMP is observed. At day 14, an increased immuoreactivity for MMP-2, MMP-9 and MT1-MMP is detected in the endothelium and mesenchymal cells (arrows). L, lumen; it, intimal thickening. Scale bars = 100 and 50 µm. (B) Western blot analysis for MMP-2 and MMP-9 in aorta extracts from days E7, E14 and E21. Extracts (50 µg of total protein) were separated by 10% SDS-PAGE under reducing conditions and immunoblotted with polyclonal antibodies against MMP-2 and MMP-9. (C) Immunolocalization of MMP-2, MMP-9 and MT1-MMP in monolayers of endothelial cells. Strong cytoplasmic reactivity for MMP-2 and MMP-9 is observed in many of the endothelial cells of the monolayer and in most of the separating, detaching, and migrating cells.
