(a) Image of a fixed HeLa cell following transient gene expression of EYFP-Sec23A using wide-field epifluorescence microscopy. The ERES are marked by the fluorescent-tagged proteins. (b) Experimental tracking of an individual ERES (highlighted by a red box) gives an estimate of the instrument precision under these conditions. The fluorescence emission from EYFP-Sec23 molecules was measured at 20.1195fps, and the location of the origin for the fluorescence was tracked in an overall sequence of 1000 frames
