The total RNA and the cell lysate (n=5 per group) were prepared from the sorted CD4⁺CD25⁻ T cells and CD4⁺CD25⁺ T_REG cells. AC9 messenger RNA was analysed by using real-time PCR (left), and AC9 protein was quantitated by densitometric analysis after Western blot (right). (B,C) The expression of miR-142-3p in CD4⁺CD25⁻ T cells and CD4⁺CD25⁺ T_REG cells. The expression of miR-142-3p was detected by (B) RT–PCR and (C) real-time RT–PCR.
