Identification of a C-mannosylation site in mindin. (A) LC-MS/MS result of C-mannosylated glycopeptides (VSLW@SSWGLC#GGPC#GK, +2, m/z 956.9320, @=162.0528 Da and #=57.0215 Da) produced by digestion of mindin with endoproteinase Glu-C and trypsin. A full FTMS spectrum at 96.54 min of liquid chromatogram has the doubly charged C-mannosylated glycopeptides with 2.1 p.p.m. mass accuracy. (B) An MS/MS spectrum shows the fragmentation of the glycopeptides for peptide identification and a cross-ring cleavage on zero and two carbons of mannose by neutral loss.
