Concentration-dependent effects of DMPO on the dimerization and formation of the protein radical-derived nitrone adducts. (A) SDS-PAGE. (B) Anti-DMPO immunostain. (C) ELISA analysis. Reactions including Mtb KatG (10 μM) and DMPO as indicated were initiated with 1 mM tert-BuOOH, and the mixtures were incubated for 1 h at 37 °C in 0.1 M phosphate buffer (pH 7.4). Reactions were stopped by the addition of 20 mM potassium cyanide. Each lane contained 1.6 μg of Mtb KatG.
