p53 C-terminal peptide (amino acids 302–390) blocks the sequestration of endogenous wt p53 at cytoplasmic sites and relocates p53 to the nucleus. The figure shows immunofluorescence of SH parental cells (AC), SH cells transfected with vector alone (D), and two independent DD1-expressing subclones, SH DD1-10 (EH) and SH DD1-8 (IL). Fluorescein isothiocyanate (FITC)-based p53 localization with C-terminus-specific PAb 421 (A, E, and I) or N-terminus-specific PAb 1801 (BD, FH, and JL) is shown. Note that PAb 421 fails to recognize sequestered p53 in the SH parental cells. Furthermore, PAb 1801 is specific for p53. (Scale bar = 10 μm.)
