αAPF stains heptameric α-hemolysin pores. A, αAPF stains the mature pore band that runs at ∼250 kDa and only weakly stains a partially misfolded protomer band at 48 kDa. In contrast, A11 weakly stains the mature pore and intensely stains the protomer bands. Neither antibody stains the 33-kDa monomer band which is the major species stained by Coomassie Blue. B, incubation of α-hemolysin with deoxycholate promotes the 250-kDa heptameric pores stained by αAPF in a time-dependent fashion.
