A. Comparison of EGFR receptors abundance on the A431, A549, and HeLa cells. Cells were plated on glass coverslips overnight, following serum starvation for 1 h the cells were immunostained with the anti-EGFR monoclonal antibody (LA22) (green) and the nuclei were stained with DAPI (blue). B. Visualization of antibody-mediated binding of CP-MVP-Z/mCherry-mINT vaults to EGFR in the indicated cell lines. Cells were plated on glass coverslips overnight. Following serum starvation for 1 h, the cells were incubated with antibody-bound vaults (red, LA22+CP-MVP-Z/mCherry-mINT) for 1 h at 4°C, fixed and the nuclei were stained with DAPI (blue). The observed red fluorescence is the intrinsic fluorescence from the mCherry protein packaged inside of the vaults.
