Expression of gene markers of alternative activation in lung macrophages by γ-herpesvirus infection. (A) Determination of FIZZ1 transcripts by quantitiative RT-PCR in lungs of mock- or MHV68-infected IFN-γR−/− mice collected at the indicated time points (in days). Data were normalized against β-actin (n = 2 per group). (B) BALF from mock-infected and infected mice collected at different time points after infection were subjected to Western blot analysis for FIZZ1. Two different animals were analyzed for each time point. Notice two peaks in FIZZ1 expression, at the acute phase and at Day 180. Blots were striped and reprobed with anti-prosurfactant B antibody. (C–D) Immunohistochemical staining for FIZZ1 in mock-infected and infected mice at Day 180, respectively. Positive staining was found in macrophages and epithelial cells from infected animals. (E) IGF-1 expression as determined by immunostaining of lungs of MHV68-infected IFN-γR−/− mice at Day 180 after infection.
