Alkylation protects Gal-1 from oxidative inactivation. A and B, mass spectrometry analysis using MALDI-TOF of Gal-1 (A) without treatment or (B) following treatment with iodoacetamide. C, Gal-1 or iGal-1 were incubated for 24 h in PBS at 37 °C followed by subjection to affinity chromatography over lactosyl-Sepharose. D, Gal-1 or iGal-1 were incubated for 24 h in PBS at 37 °C followed by addition of 20 μm of each galectin to HL60 cells for 4 h. Cells were then stained with annexin-V fluorescein isothiocyanate and propidium iodide and analyzed for PS positivity by flow cytometric analysis.
