MitoTEMPOL protects mitochondrial DNA from damage by menadione. (A–C) C2C12 cells were seeded in 6-well plates at 300 000 cells/well and left to attach overnight in medium. They were then pre-incubated for 1 h with medium containing 50 μM MitoTEMPOL or propylTPP after which 25 μM menadione was added and cells were incubated for a further 1 h at 37°C. Cells were then harvested by scraping, the DNA was isolated and amplified by QPCR and the product quantified using PicoGreen.
