pH dependence of metal binding by Apo-MnSOD. Co2+ uptake was monitored fluorimetrically as described in the Methods. MnSOD (50 ug/mL) in 2.5 mL sample buffer was equilibrated at 37°C for 10 min, and the reaction initiated by addition of 24 μM CoCl2 (approximately 10 stoichiometric equivalents). The pH of each sample was measured directly in the reaction cuvette. (Top) pH dependence of the fast phase gating fraction (A0/ATOT). A fit of the data to a modified Henderson-Hasselbalch equation as described in the text is shown (pKa = 7.7). (Bottom) pH dependence of the slow phase rate constant, kslow.
