SW620 cells were cotransfected with human TLR2 and TLR1 (the latter in wild-type or point mutant versions, as indicated), as well as with a firefly luciferase reporter gene and a Renilla transfection control. After 48 h, the cells were stimulated for 6h with LT-IIb-B5 (2 μg/ml) (A) or Pam3CSK4 (20 ng/ml) (B). Cellular activation is reported as relative luciferase activity, normalized to that of cells transfected with reporter and empty vectors. Results are presented as means ± SD (n = 3). Asterisks indicate significantly (p < 0.05) reduced activities compared to transfectants that received wild-type (WT) TLR1.
