EGFP-fused ORF45 expression vectors were transfected into 293T cells. Green fluorescence was visualized 24 h after transfection. As indicated, some transfected cells were treated with 10 ng/ml LMB, which specifically inhibits CRM1-dependent nuclear export. (B) HA-tagged ORF45 expression vectors were transfected into 293T cells. The transfected cells were fixed, permeabilized, and immunofluorescently stained with anti-HA antibodies (red). The cells were also counterstained with DAPI to show the nucleus (blue). (C) KSHV ORF45 rapidly accumulates in the nucleus after LMB treatment.
