Cytokine production of memory T cells is influenced by tissue localization. Lymphocytes from spleen and ILN were isolated at day 80 and after 5 h ± peptide stimulation stained with Thy 1.1, CD8, IFN-γ, and TNF-α. Gating on Thy 1.1+ CD8+ cells, stimulation with GP33–41 specific peptide results in IFN-γ and TNF-α. No staining of IFN-γ or TNF-α was seen with no peptide controls (data not shown).
