Dye incorporation is strongly dependent on NaBH4 reduction. A total of 400 pmol of either tRNAPhe(red) or untreated tRNAPhe was treated with Cy3 using the standard labeling protocol (Materials and Methods). Peaks at 260 and 550 nm correspond to the absorption of tRNAPhe and Cy3, respectively.
