HIF-2α PAS-B selectively binds to conformation 1 of the ARNT PAS-B Y456T point mutant. Sections of 15N-1H HSQC spectra of a titration of unlabeled HIF-2α PAS-B (red, 0 μM; orange, 40 μM; gold, 100 μM; blue, 200 μM; violet, 950 μM HIF) into 75 μM 15N-labeled ARNT-PAS B Y456T are shown. Peaks are labeled with conformation (1 or 2) and residue name/number. The wild-type conformation 1 of Y456T still binds HIF-2α as shown by HIF-dependent chemical shift and intensity changes, whereas signals from the new conformation 2 are unaffected.
