Estrogen inhibited ischemia-induced FADD expression in OVX mice. Levels of FADD mRNA and protein were measured with RT-PCR (A and B) or Western blot (C), respectively. Final results were normalized to GAPDH levels. (A) FADD mRNA levels were significantly higher in ischemic cortex of OVX mice at 3, 6 and 12 h reperfusion compared to contralateral cortex (control) as well as those in sham-treated mice without MCAO. (B) Estrogen replacement attenuates ischemia-upregulated FADD mRNA in ipsilateral cortex of OVX mice at 6 h reperfusion. (C) Estrogen-replaced mice exhibited significantly lower FADD protein levels in ipsilateral cortex compared to OVX mice at 6 and 12 h reperfusion. *versus control; **versus non-treated OVX mice (p<0.05).
