Kinetics of activation of TAFI variants by thrombin alone. TAFI-WT (•), TAFI-K42/43/44A (○), TAFI-K133A (▾), TAFI-K211A (▿), TAFI-K212A (▪), TAFI-R220A (□), TAFI-K240A (♦), and TAFI-R275A (⋄) were incubated at various concentrations with IIa. The rate of TAFIa formation (mol of TAFIa formed/mol of thrombin/second) was calculated. The slope of the relationship between the rate and TAFI concentration was measured to determine the catalytic efficiency (kcat/Km1) of TAFI activation.
