(A) Data from flow cytometry analysis show the expression of CD41, CD45, c-Kit, and CD34 in EB6 cells. The sorting gates for CD41⁻ or CD41⁺ cells, c-Kit⁺CD41⁺ or c-Kit⁻CD41⁺ cells, and CD34⁺ CD41⁺ or CD34⁻CD41⁺ cells are indicated as squares. (B) EB6 cells were fractionated based on expression of CD41, c-Kit, and CD34, co-cultured with OP9 cells for 4 days, sorted for GFP+ cells, and transplanted into lethally irradiated mice. HOXB4 expression was maintained from in vitro co-culture through in vivo repopulation. Recipient mice were analyzed 16 weeks after transplantation. Over 95% of reconstituted blood cells were of myeloid lineage in all cases (data not shown). Two independent experiments gave similar results. Data from one experiment are shown.
