(A) Immunoblot analysis of extracts harvested from HTC75 cells 48 h after double infection with lentiviral tankyrase 1 shRNA and GFP shRNA or two different lentiviral TIN2 shRNAs. (B) Quantification of mitotic cells with persistent sister telomere associations as measured by chromosome-specific telomere FISH using a subtelomeric 16pter probe. Approximately 100 mitotic cells were scored for each sample. (C) Quantification of the frequency of sister chromatid fusions as measured by telomere PNA FISH. 25 metaphase spreads were scored for each sample. (D) Quantification of SA β-galactosidase–positive cells. Approximately 500 cells were scored for each sample. (E) Quantification of BrdU-positive cells.
