Transcriptional analysis of ycnK- and csoR-dependent ycnJ expression. Two micrograms of total RNA isolated from the WT and mutants under normal, 0.25 mM BCS, and 0.5 mM copper excess conditions was blotted onto a nylon membrane by dot blotting and hybridized with a digoxigenin-UTP-labeled antisense riboprobe specific for the ycnJ transcript. Hybridization signals were detected by using a digoxigenin-specific antibody fragment conjugated with alkaline phosphatase and AttoPhos as chemifluorescence substrate.
