US3-dependent phosphorylation of gB depends upon the kinase activity of US3. HaCaT cells were infected with HSV vRR1205, which expresses kinase-inactive US3 (K220A), vRR1202 (US3 mutant), or US3-R for 6 h. Subsequently, the cells were labeled for 6 h with either [35S]methionine-cysteine (right) or 32PO4 (left). Cells were extracted with detergents and not fractionated as in Fig. 1. gB, gH, and gE were immunoprecipitated from detergent extracts of membranes.
