Table 2.

Temperature dependence of the observed binding enthalpy (ΔHob) and calculated heat capacity change (ΔCp) for the 1,8-naphthyridine binding to cytosine in the 21-meric AP site-containing DNA duplex

ΔHobs (kcal/mol)a
ΔCp (cal/mol K)b
20°C 15°C 10°C 5°C
AND −20.5 ± 0.6*1 −20.0 ± 0.6*2 −19.1 ± 0.5*2 −18.2 ± 0.1*2 −161
ATMND −12.8 ± 0.7*2 −11.7 ± 0.5*2 −10.6 ± 0.1*1 −9.5 ± 0.2*2 −217

aΔHobs was directly determined by ITC experiments. Errors are the standard deviations obtained from at least three independent measurements at each temperature (*1 4 times; *2 3 times). Sample solutions were buffered to pH 7.0 with 10 mM sodium cacodylate, containing 100 mM NaCl and 1.0 mM EDTA. DNA duplex: 5′-GCA GCT CCC GXG GTC TCC TCG-3′/3′-CGT CGA GGG CCC CAG AGG AGC-5′, X = AP site (dSpacer), C = target cytosine.

bHeat capacity change calculated from the slope δ(ΔH)/δT obtained by linear least squares fit (cf. Figure 6, r = 0.9906 for AND, r = 0.9999 for ATMND).
