A. GFP-tagged Ura3-2 was imaged in cells incubated at 25°C or 30 mins after a shift to 37°C, in the presence or absence of cycloheximide. B. GFP-tagged Ura3-2 was expressed from the GAL promoter, then expression repressed by growth in glucose prior to temperature shift, in the absence of cycloheximide. Degradation was normal even though the aggregation state of the protein was different from that in cycloheximide. C. GFP-tagged lambda repressor in cells gown at 16°C, or one hour after shifting to 30°C. The images have been approximately normalised for intensity.
