Vegetative B. subtilis PS533 cells were prepared, stained and analyzed by flow cytometry as described in Methods.

Peak fluorescence values in arbitrary units were determined from the position of the major peak (generally > 85% of all cells) of fluorescent cells in flow cytometry. All values are averages of at least duplicate experiments and individual values differed by < 25% from the averages.

Cells were heated for 30 min at 80°C before or after DiOC6(3) addition.
