Inhibition of purified 20S proteasomes by organotins. Try, trypsin. Potency of phenyltins (A) and butyltins (B) to inhibit CT-like activity of purified 20S rabbit proteasomes. (C) TPT preferably inhibits CT-like activity of purified 20S rabbit proteasome. (D and E) Purified 20S proteasomes (4 μg) were incubated with 10 μmol/L of TPT or the solvent methanol (Sol). After removing the unbound TPT from the proteasomes in an Ultrafree-MC centrifugal filter unit (MW cutoff, 5 kDa), the proteasome preparations were measured for CT-like activity (D) and β5 subunit levels in Western blotting (E). (F) TPT at 10 μmol/L inhibited proteasomal CT-like activity in a time-dependent manner. Data are mean ± SD from three experiments.
