Effect of glycated albumin on PKCα and Src activation in
vivo. 4-Week-old female mice of the C57/BL6 strain were injected with
50 mg/kg of HGA or HA or PBS either directly in the quadriceps
(GA-Im: a, b, c, and d) or in the tail vein
(GA-Iv: e, f, g, and h) as described under
“Experimental Procedures. a and c, muscle tissue
lysates from muscle-injected mice were blotted with anti-phospho PKCα,
and PKCα phosphorylation was analyzed by densitometry. b and
d, muscle tissue lysates from muscle-injected mice were blotted with
anti-phospho Src Tyr416 antibody and Src phosphorylation was
analyzed by densitometry. e and g, muscle tissue lysates
from tail vein-injected mice were blotted with anti-phospho PKCα, and
PKCα phosphorylation was analyzed by densitometry. f and
h, muscle tissue lysates from tail vein-injected mice were blotted
with anti-phospho-Src Tyr416 antibody, and Src phosphorylation was
analyzed by densitometry. A representative autoradiograph is shown.
