Total RNA was extracted from the indicated cells that had been treated with VacA (20 μg/ml) for the indicated times and used for RT-PCR. M, size marker. (B) Jurkat cells were incubated with VacA for the indicated times. Cell lysates were prepared at the indicated incubation times and subjected to immunoblotting with the indicated antibodies. (C) Effects of PI3K and p38 MAP kinase inhibitors on VacA-mediated IL-12 p40 expression in Jurkat cells. Cells were pretreated with LY294002 (6.25 μM) or SB203580 (6.25 μM) for 1 h, followed by VacA treatment for 2 h, and then IL-12 p40 mRNA expression was analyzed by RT-PCR.
