FGF-2 induces intracellular relocalization of the PKA catalytic subunit
to the nuclear membranes and into the nucleus. (a)
Quantification of PKAC in nuclear proteins of
FGF-2-stimulated NIH 3T3 cells by Western blot analysis. Serum-starved
NIH 3T3 cells were treated with FGF-2 (10 ng/ml) or PKA activator
8-Br-cAMP (200 μM) for 1 hr, and nuclear proteins were isolated. Ten
micrograms of nuclear proteins was separated in 10% SDS/PAGE,
blotted onto a nitrocellulose filter, and detected with
anti-PKAC antibody. (b) Detection of
PKAC in FGF-2 stimulated NIH 3T3 cells by indirect
immunofluorescence confocal microscopy. Serum-starved NIH 3T3 cells
were treated with FGF-2 or PKA-activator 8-Br-cAMP (200 μM) for 1 hr,
fixed, and stained with anti-PKAC.
