Table IV.

Comparison of glutathione-conjugate transport capacity in crude membrane vesicles prepared from human erythroleukemia K562 cells and by RLIP76 purified from K562 cells and transformed E. coli.

Fraction 3H-DNPSG transport 3H-GSHNE transport IC50 for 4-HNE
K562 cells purified RLIP76 39.4±2.6 nmol/min/mg 4.2±0.3 nmol/min/mg
Recombinant purified RLIP76 125±14 nmol/min/mg 4.8±0.4 nmol/min/mg
IOVs from:
Control K562 cells 147±11 pmol/min/mg 51 ±4 pmol/min/mg 1.8±0.2μM
Vector-transfected K562 cells 166±18 pmol/min/mg 56±6 pmol/min/mg 2.0±0.2μM
RLIP76-transfected K562 cells 738±42 pmol/min/mg 294±31 pmol/min/mg 9.4±0.6μM

Inside-out vesicles were prepared from K562 cells according to the protocol as described by us previously (8). RLIP76 was purified by DNP-SG affinity column chromatography from K562 cells as well as from recombinant source and reconstituted into artificial liposomes to perform transport studies.
