Gel filtration chromatography of the Alg14p membrane subunit of the ER UDP-GlcNAc transferase. Detergent extracts from cells expressing HA-ALG14 (XGY151) (Gao et al. 2005) were prepared in the presence of 0.1% Triton X-100 and fractionated over a Superose 12 column (see Materials and methods). An aliquot of each fraction was subjected to SDS–PAGE and analyzed by immunoblotting with the anti-HA-antibody. Molecular weight protein calibration markers, whose fractionation profile is graphed in the upper panel, were fractionated in parallel under identical conditions.
