Role for Hey1 in suppression of adipogenesis by FOXC2. A, 3T3-L1 preadipocytes were infected with a retrovirus vector alone (pBabe or pLNCX2) or retroviruses carrying the genes for FOXC2-VP16, FOXC2-Eng, or FOXC2 as indicated. Two days post-confluence, the cells were induced to differentiate with MDI. RNA was purified at the indicated times and expression of Hey1 analyzed by quantitative PCR. B, 3T3-L1 preadipocytes were infected with a retrovirus vector alone (pBabe) or a vector carrying the gene for Hey1. Two days after confluence, the cells were induced to differentiate in presence of MDI or MDI+T. The cells were stained with Oil Red-O on day 14. C, on the indicated days of differentiation, the cells were lysed, and expression of C/EBPβ, C/EBPα, PPARγ, and FABP4 was analyzed by immunoblot analysis.
