Protein-DNA interactions were performed in 20-μl reactions containing the indicated concentration of MutS proteins and 5 nm oligonucleotide duplex. The reactions were incubated on ice for 20 min, and the products were analyzed by gel shift assays. Representative gel shift analyses for MutSα and MutSβ are shown in B and D, respectively, and the relative binding activities of individual reactions determined using the average binding value of two independent experiments are plotted in C and E.
