Interaction of PCNA peptides with RFC. During a 7-min interval, a solution of 0.1 mg/ml RFC in 35 μl of buffer (10 mM Hepes⋅HCl, pH 7.4) passed over the surface of streptavidin-coated BIAcore sensor chip to which each of 50 overlapping biotinylated PCNA peptides were immobilized at a density of 100 resonance units. After completion of this 7-min injection step, buffer was passed over each chip at the same flow rate (5 μl/ml). The data presented here indicated the binding (in resonance units) of RFC to PCNA peptides containing residues 36–55 (A), 196–215 (B), and 166–185 (C). The binding pattern observed with PCNA peptide 166–185 was identical to that obtained when a control chip devoid of PCNA peptides was used.
