J774A.1 macrophages (A,B) or HEK293 cells transfected as indicated (C) were pretreated or not with MCD (10 mM) or cytochalasin D (5 µg/ml). Half of the MCD-treated groups were subsequently reconstituted with cholesterol (1 mM). The cells were stimulated or not with P. gingivalis (MOI = 10:1) and assayed for TNF-α (A) or IL-6 (B) by ELISA, or for NF-κB activation (C) reported as relative luciferase activity (RLA). Data are means ± SD (n = 3) from one of two sets of experiments with similar results. Asterisks show statistically significant (p < 0.05) inhibition of cell activation.
