Kinetics of rhodopsin formation by using purified WT and mutant opsins. WT opsin and mutant opsins at positions W265 and A164V were prepared in buffer containing CHAPS [1%(wt/vol)] and DMPC [1%(wt/vol)], as described in Methods. Opsins at about 0.7 μM concentrations in the mixtures were treated with 11-cis-retinal and increases in absorbance at 500 nm were measured as a function of time.
