Partitioning of Cx43 connexons into the TX-114 aqueous phase is sensitive to the folding status of Cx43. NRK cells were labeled for 4 h at 20°C. A TX-114 aqueous (connexon-containing) fraction was prepared and cross-linked with EGS. One-half of the sample was incubated without (control), and the other half with, 1% N-lauroyl sarcosine (N-LS) in the presence of 20 mM DTT. After detergent exchange, the samples were subjected to TX-114 phase partitioning at 30°C before immunoprecipitation of Cx43. See Materials and Methods for details.
