Chondrocytes in serum-free medium were pre-treated with or without 10 uM JAK-3 inhibitor for 45 minutes followed by stimulation with 10 ng/ml IL-7 for 16–18 hr. Conditioned media obtained after the incubation period was analyzed for S100A4 by immunoblotting and for MMP-2 as a loading control.
