CE accumulation in acLDL-loaded macrophages is repressed during ACAT inhibition, which dominates FC accumulation. To quantify intracellular storage of cholesterol, THP-1 macrophages were incubated for 48 h in RPMI medium containing 10% LPDS with or without 100 µg/ml of acLDL and the indicated concentrations of OAA. For quantification of secreted cholesterol, the cells were washed extensively and incubated for a further 24 h without acLDL. (A) Cells were fixed with 4% paraformaldehyde and stained with oil red O.
